We were developing a sitcom one time and the producer - an otherwise ineffective buffoon who may be reading this - pointed out that, often in Seinfeld, the most minor of characters would have a tick or a quirk to make them a little more interesting. I went through the script and, in the case of one character, randomly capitalised words in his speeches. When it came to the performance, the actor playing him sort of shouted or blurted out those words. It was very funny and kind of sounded like he was either very tense or on the verge of belching. Think of traits for your smaller characters.
