Don't write Episode One. It can ensnare a writer into exposition and introductions that are unnecessary and unusable in any other episode - so, you may as well crack how you're going to set the scene and introduce the characters in every other episode that isn't the first.
Make sure the humour is driven by the characters and stories, and not just people saying funny lines you've put into their mouths. Writing comedy for radio is so much more than gag writing. It's a good idea to limit the number of formulaic lines that begin 'That's like a cross between...' or 'That's about as healthy as...' or 'I haven't seen anything as bad as that since...'
