The annual show, which began in 1912 when King George V and Queen Mary attended, this year will honour the life of Queen Elizabeth II. It will be recorded at the Royal Albert Hall on 1st December, for broadcast on ITV later in the month.
The Not Going Out star, who has previously presented the Queen's Platinum Jubilee concert in June, joked: "I was actually going to buy tickets for this year's Royal Variety Performance, so not only is it a real honour to be asked to host it, it's also saved me a few quid.
"I now just have to work out the logistics of introducing these brilliant acts and also rushing back to my seat to watch them."
The comedians set to perform as part of the night are Al Murray as The Pub Landlord, Maisie Adam, Omid Djalili and Britain's Got Talent winner Axel Blake.
Meanwhile Frank Skinner, David Baddiel and The Lightning Seeds will present a new version of football song Three Lions, which is being re-written as a Christmas track to reflect the Lionesses' Euro 2022 win and the Qatar World Cup.
The Queen will be honoured in a segment organised by Andrew Lloyd Webber, Gary Barlow, Gareth Malone and the London Youth Choir.
Music stars set to perform as part of the show include Nile Rodgers & Chic, George Ezra, Ellie Goulding, Sam Ryder and Becky Hill. Stage shows presented will include Cabaret, Newsies and Cirque Du Soleil.
Giles Cooper, chairman of the Royal Variety Charity, says: "The support from ticket sales for the performance means that the Royal Variety Charity continues to thrive, even given the current economic climate being felt by so many throughout the charitable world."
