How would you go about it?

We've got a sitcom with the pilot episode ready to film once we've got all the actors sorted.

Once filmed, what's the best route to take with it?

Well, if you're actually going to film it rather than send the script out, just put it somewhere online, either public or private, and send the links to producers and people in the business. You could get fancy and send out DVD copies, but it won't make much of a difference.
