The things I found most helpful were
1. To get a good line, write 10
2. Start a sitcom in Episode 3
3. If it's a dramatic situation it can almost always be a comic situation.
4. Talk through plot with someone else.
5. The problem with comedy that's not working is usually either you don't care or you don't believe what's happening.
I know most of these are cliches of writing courses, but I still think they're useful when you're staring at a blank page.
They seemed like two nice blokes too.
I liked:
"make the funny bits longer and the less funny bits shorter"
although they admit that's Richard Curtis' tip, not theirs.
