It's geeky, I know, but I have recently been enjoying all 681 episodes of Order of the Stick.

Maybe the title of this thread should be changed? It is online, true, but it isn't funny.

I don't play online games but even I find this series well done and funny: The Guild
By the 2nd season the characters who I found completely annoying in season 1 were my favourite.
I advise watching them in order starting with season 1 episode 1.
Season 3 is coming out soon too. The show got so popular Microsoft sponsored it and you can buy the season on DVD.

Plus the readhead is nerdelicious! hot!
EDITED: Just found out season 3 already has a few episodes out. They've just moved to Microsoft's website. You can access them through the website http://www.watchtheguild.com/
